Melatonin in Skincare: Science or Marketing?

Melatonin, often known as the sleep hormone, is making waves beyond the realm of sleep aids. Recently, it has started appearing in serums and facial creams, promising to protect the skin from premature aging. But what’s the science behind these claims? Current research suggests that the potential of melatonin may be greater than many realize.

The Skin’s Own Production of Melatonin

Traditionally associated with regulating the sleep-wake cycle, melatonin is less commonly recognized as a substance produced by our skin. Skin cells possess their own melatonin receptors and can create enzymes responsible for its synthesis and degradation. In this way, the skin has an independent melatonin system that functions apart from sleep. Research by Kleszczynski and Fischer in 2012 pointed out melatonin’s protective role in the skin. As a potent antioxidant, it captures free radicals produced by UV radiation, protects mitochondria, and can limit oxidative cell damage—a significant factor in photoaging.

Protection Against Aging Rather Than Just Wrinkle Reduction

Those expecting melatonin to function like retinol—famous for its wrinkle-smoothing properties—might be disappointed. Current scientific understanding indicates that while melatonin does not significantly stimulate collagen production or directly smooth out existing wrinkles, it plays a protective role. Doris Day and colleagues summarized the state of research in 2018 in the Journal of Drugs in Dermatology, highlighting that topically applied melatonin can neutralize free radicals and activate antioxidant enzymes like superoxide dismutase and catalase. This means the skin could become more resilient to UV-induced oxidative stress. However, clinical studies confirming these anti-aging effects are still lacking.

Promising Initial Clinical Data

Even though biological effects of melatonin haven’t been extensively documented, a clinical study by Milani and Sparavigna published in 2018 in Cosmetics examined a melatonin-containing facial cream over several weeks. Participants reported improved skin hydration, a smoother skin surface, and slight reductions in fine lines. Nonetheless, the study had limitations—it was small, non-placebo-controlled, and included authors affiliated with the product’s manufacturer. While the results provide intriguing insights, they do not serve as conclusive proof of efficacy.

Melatonin may play a role not only in protecting against skin aging but also in preventing certain skin cancers. In cellular experiments, research teams have shown that melatonin exhibits a stronger antioxidant effect than vitamin E or C. According to Professor Hendrik Fischer, melatonin might be especially significant for organ transplant recipients who must take immunosuppressants, increasing their risk of skin cancers.

Research Points to Potential

A more recent review by Bocheva and colleagues in 2022, published in the International Journal of Molecular Sciences, concluded that melatonin and its metabolites protect skin cells from oxidative stress, reduce inflammatory reactions, and lower UV-induced DNA damage. There is also evidence suggesting that melatonin might indirectly slow the breakdown of collagen by reducing the activity of certain enzymes, called matrix metalloproteinases.

Further, Slominski and colleagues, in a review set to be published in the Journal of Investigative Dermatology in 2025, label melatonin as a crucial component of the skin’s natural defense system. However, they also stress the need for larger, placebo-controlled studies over extended periods before definitive claims regarding its anti-aging effects can be made.

Conclusion: A Matter of Potential and Further Research

While the emergence of melatonin in skincare is intriguing and supported by some scientific evidence, a gap remains between potential benefits and proven efficacy. As more research unfolds, the beauty industry may need to temper its marketing promises and focus on robust data to affirm melatonin’s role in skincare. Until then, it might be wise to view melatonin-enriched products as part of a broader skincare regimen rather than a magic solution for anti-aging.
